Movie S3. Increased T-cell motility and decreased T cell–CD11c+ cell interactions in a heavily infiltrated islet. A combination of 10% CD2-dsRed.OT-I CD8+ T cells and 90% OT-I CD8+ T cells were transferred into RIP-mOva.CD11c-YFP recipient mice. Islets were isolated 7 d following T-cell transfer and immobilized in low-melting agarose. Isolated islets were imaged by time-lapse two-photon microscopy at 30-s intervals for 30 min. Representative time-lapse imaging of highly motile T cells in a heavily infiltrated islet. (Scale bar, 20 μm.) Z depth of 12 μm; time stamp, min:sec.
