Movie S7. Three-dimensional reconstruction of confocal stacks taken at the optic nerve head of a glutamate transporter 1 (GLT1)::EGFP mouse labeled by TUNEL to detect degrading DNA and by MitoFISH to detect mtDNA. Similar imaging parameters were used as in Movies S5 and S6. DAPI is shown in blue, GFP is shown in green, MitoFISH is shown in red, and TUNEL is shown in white. GFP and MitoFISH are shown individually, and MitoFISH is then overlaid with TUNEL to visualize volumes of overlap. Volumes of overlap (TUNEL-positive and mFISH-positive voxels) were localized by Imaris (magenta) and flashed onto the MitoFISH and TUNEL data. The TUNEL-positive, MitoFISH-positive signal is then covisualized with GFP to show that mitochondrial degradation occurs within astrocytes. Finally, the raw GFP, MitoFISH, and TUNEL signals are all shown together.
