Expression of C3 inhibits AMPH-induced DAT internalization. SK-N-SH cells were cotransfected with mRed-DAT and C3-GFP. The cell in the lower left expresses both C3 and DAT, and the cell in the upper right expresses DAT alone (see also Fig. 1C, which shows a single frame from the movie). AMPH fails to produce clustering or internalization in the cell expressing C3 but it does produce robust clustering and internalization in the cell expressing DAT alone. AMPH also stimulates lamellipodia formation in the cell expressing DAT alone, consistent with Rho/Rac activation.
