Alanine mutations in the C-terminal MT-binding site of KHC (KHCmutA) inhibit MT sliding in S2 cells. Representative photoconversion time-lapse movies of S2 cells transfected with tdEOS-tubulin and wild-type KHC (Left) or KHCmutA (Right). Endogenous KHC was depleted in both cells by dsRNA against the 3′-UTR. A small population of MTs was photoconverted and imaged for 9 min. Note that KHCmutA slides MTs much slower than wild-type KHC.
