Three-dimensional reconstruction of a “multicolor” mouse model that allows the visualization of the different cellular compartments in tumors. In the experiment from Fig. S7 B–D, a Z stack was generated from a 22-d MC57-Cerulean tumor by acquiring 11 images (optical slices) at 5.97-μm intervals (total depth of 66 μm). A 3D reconstruction of the resulting 66-μm-thick tumor section was generated using Imaris. The blue regions (cancer cells) fade away to better visualize the stromal compartments.
