R848 does not induce endothelial cell death. 3D reconstruction of mouse mesenteric veins obtained by confocal intravital microscopy. Two hours after local stimulation of mesenteric vessels with R848 (100 µg), nuclei of dead cells (blue) and the luminal side of the endothelium (green) were detected by i.v. injection of Sytox Blue (10 μL, stock 5 mM) and AF488-conjugated anti-PECAM1 (CD31, 10 μg), respectively. No dead endothelial cells were observed. The small tick bar indicates 5 μm.
