Movie S2.

A video showing cryo-FIB-SEM serial sections that were aligned to form the same data cube of X = 20 µm, Y = 12 µm, Z = 11 µm, with a voxel size of 10 × 10 × 20 nm as in Movie S1. Segmentation of the reconstructed 3D is shown for several vesicles and a part of the plasma membrane. The purple color shows the outline of the plasma membrane of a PMC. In the continuation of the video, the purple color cumulatively designates the plasma membrane in 3D and this then reveals a complex of connected vesicles (light purple) on the inside of the cell that has openings through the plasma membrane.
