Activation of a sarcomere with SLi very close to contractile filaments overlap limit (∼3.5 μm). This experiment shows that it is still possible to fully activate a sarcomere using a microperfusion, even in conditions starting at very long SLi. Note that such behavior is not observed when a myofibril is completely activated (lower part of Movie S6).
