Volume projection of a particle where integrity was lost measured by live-cell imaging of MDMs infected by dual-labeled particles and unlabeled simian immunodeficiency virus (SIVmac) VLPs carrying Vpx. The sample was fixed 50 min after initial imaging time, counterstained with DAPI (blue), and stained for p24/p27 (green). The remaining IN-mRuby signal in the viral complex with 3D-SIM allows the observation that the integrity loss of the capsid occurred in the cytoplasm and the given particle retains CA. Several SIVmac VLPs that delivered Vpx are also detected by the p24 staining (this projection corresponds to Fig. 6D).
