Movie S4.

Time-lapse image of a centered, dual-labeled HIV particle during infection. Plotting of the measured mean intensities with background subtracted (y axis) within the voxels of the tracked individual particle is shown. (Top Left) Two drops of GFP signal are discerned in the presence of Vpr. (Bottom Left) Particle boxes show the signal of GFP (green) and Vpr (red) over time. (Bottom Left and Right) Other boxes show the signal (gray) of each individual fluorescence measurement as labeled. During time-lapse imaging, a brightfield reference image was taken for every time point (gray) and mixed with z-projected fluorescence acquisitions; only one particle fused into the cell that produced de novo HIV-Gag-iGFP.

Early cytoplasmic uncoating is associated with infectivity of HIV-1
