Inactivation of RapR-Src disrupts relocalization of PH-AKT in HeLa cells. A HeLa cell infected with adenovirus expressing RapR-Src-as2-cerulean-myc and transfected with PH-AKT-mVenus and iPEP-mCherry-FRB constructs (transient transfection) was imaged for 2 h every 2 min. Rapamycin (500 nM) was added after 30 min (indicated by appearance of “Rapamycin Added”), and DMSO was added after 15 min (indicated by appearance of “1NA-PP1 Added”). The movie shows pseudocolored images of PH-AKT-mVenus. Warmer colors reflect higher local fluorescence signal. Nucleus is masked to clarify perinuclear/peripheral distribution of PH-AKT.
