Representative movie of mixed populations of NRVMs expressing the FRET-based PKA phosphorylation-sensitive sensor AKAR4 or its mitochondrial targeted version OMM-AKAR4. Cells were treated with low Fsk (20 nM) and subsequently CsA 200nM and CalA 50nM (to inhibit phosphatases). Final treatment with Fsk 20μM combined to IBMX 100 μM saturated both sensors. Scale: Cold shades (low PKA-dependent phosphorylation) hot shades (high PKA-dependent phosphorylation).
